ABSTRACT

BACKGROUND:Chemotherapy drugs have been shown to penetrate the blood-brain barrier and cause neurotoxicity, further damaging nerve cells.OBEJCTIVE:To investigate the changes in the expression and morphology of hippocampal neurons in chemobrain, and to investigate the underlying mechanism of nerve regeneration in chemobrain.METHODS:The brain specimens were obtained from the six patients undergoing cancer chemotherapy and six patients without chemotherapy, and were then subdivided into two groups:< 60 years and > 60 years groups (n=3 per group).Immunofluorescence staining with specific antibodies was used to detect the changes in the morphology and number of neurons in the hippocampus CA1,CA2 and CA3.RESULTS AND CONCLUSION:The number of neurons positive for NeuN,DCX and PV in the hippocampal CA1,CA2 and CA3 in the chemotherapy group was significantly lower than that in the non-chemotherapy group. Moreover, the neurons positive for PV were smaller, with less and shorter nervous processes.Immunofiuorescence staining findings showed that the < 60 years old patients in the chemotherapy group had less neurons positive for NeuN and DCX than that in the non-chemotherapy;for > 60 years old patients, there was no significant difference between two groups. These findings suggest that chemotherapy can downregulate the expression of immature neurons in the hippocampus and reduce the number of neurons in the hippocampus. Moreover, chemotherapy can change the morphology and reduce the number of PV neurons.

ABSTRACT

Bronchoalveolar lavage is an important animal experimental technique in the study of respiratory system and its pathological changes. It can acquire a variety of biochemical factors, inflammatory mediators and immune cells from the respiratory tract and lungs, and provides an important evaluation index and reference for animal experiment. Bronchoalveolar lavage is an effective and reliable method for the diagnosis of respiratory diseases. It has been gradually standardized and widely used in clinical practice at present, however, there is no set of standard for bronchoalveolar lavage in rats and mice. The results of bronchoalveolar lavage fluid are affected by many factors, such as the lavage fluid, suction pressure, the amount of lavage and recovery, and the retention time of lavage fluid in the lungs. Successful and efficient acquisition of lavage specimens is the key to the study and evaluation of respiratory diseases. This paper summarizes the current lavage methods commonly used by domestic and foreign researchers, and provides a reference for further research in the this field.

ABSTRACT

Background Misfolding of Myocilin protein plays an important role in the pathogenesis of primary open angle glaucoma (POAG).GRP78 can transfer the misfolded proteins out endocytoplasmic reticulum and keep the protein synthesis function of endoplasmic reticulum. However the relationship between GRP78 and Myocilin in the pathogenesis of POAG has not been elucidated. Objective This study was to investigate and analyze the coexpressions of GRP78 and Myolicin in trabecular meshwork cells (TMCs) of POAG.Methods The trabecular meshwork tissues were obtained during the surgery of POAG and healthy donor for the isolated and in vitro culture of TMCs,and the morphology of the cells was compared between POAG-derived TMCs and donor-derived TMCs.The expression of specific factors for TMCs were detected by immunochemistry. The cells were divided into normal control group, tunicamycin (Tm)-treated group and staurosporine (STS)-treated group, and the cells were cultured by regular medium, the 1 μ mol/L Tm medium or 0.1 μmol/L STS medium for 24 hours respectively. The co-expression of GRP78 and Myocilin in the cells were detected using immunofluorescence assay. This study was approved by Ethics Committee of Xi'an No.4 Hospital, and written informed consent was obtained from each patient before surgery. Results Primarily cultured cells showed the fiat star-like and irregular appearance with 3-5 processes, round nuclei, abundant cytoplasm and black engulf particles. The 3-7 generations of POAG-derived cells grew well, showing positive expressions of laminin (LN),fibronectin (FN),neuronal specific enolase (NSE) and Vimentin.Immunofluorescence assay showed positive expressions of GRP78 and Myocilin in the cells of the Tm group,STS group and normal control group, with the reddish and green fluorescence separately.Incomplete co-expression of GRP78 and Myocilin was seen in donor-derived TMCs,and complete co-expression of GRP78 and Myocilin was found in the POAG-derived TMCs in the normal control group.Complete co-expression of GRP78 and Myocilin was exhibited in both donor-and POAG-derived TMCs in the Tm group and STS group. In addition, accumulation of GRP78 protein around nucleus was seen in both donor-and POAG-derived TMCs in the Tm group. Conclusions GRP78 and Myocilin proteins are completely co-expressed in POAG-derived TMCs,inferring that GRP78 and Myocilin play an interaction role in the pathogenesis and development of POAG.GRP78 may play a cytoprotective role against the apoptosis of TMCs caused by endoplasmic reticulum stress.

ABSTRACT

<p><b>BACKGROUND</b>Our previous study has confirmed that one bout of exhaustion (Ex) can cause hippocampus neurocyte damage, excessive apoptosis, and dysfunction. Its initial reason is intracellular calcium overload in hippocampus triggered by N-methyl-D-aspartic acid receptor (NMDAR) over-activation. NMDAR activation can be suppressed by γ-aminobutyric acid (A) receptor (GABAAR). Whether GABAAR can prevent intense exercise-induced hippocampus apoptosis, damage, or dysfunction will be studied in this study.</p><p><b>METHODS</b>According to dose test, rats were randomly divided into control (Con), Ex, muscimol (MUS, 0.1 mg/kg) and bicuculline (BIC, 0.5 mg/kg) groups, then all rats underwent once swimming Ex except ones in Con group only underwent training. Intracellular free calcium concentration ([Ca2+]i) was measured by Fura-2-acetoxymethyl ester; glial librillary acidic protein (GFAP) and synaptophysin (SYP) immunofluorescence were also performed; apoptosis were displayed by dUTP nick end labeling (TUNEL) stain; endoplasmic reticulum stress-induced apoptosis pathway was detected by Western blotting analysis; Morris water maze was used to detect learning ability and spatial memory.</p><p><b>RESULTS</b>The appropriate dose was 0.1 mg/kg for MUS and 0.5 mg/kg for BIC. Ex group showed significantly increased [Ca2+]i and astrogliosis; TUNEL positive cells and levels of GFAP, B cell lymphoma-2 (Bcl-2) associated X protein (Bax), caspase-3, caspase-12 cleavage, CCAAT/enhancer binding protein homologous protein (CHOP), and p-Jun amino-terminal kinase (p-JNK) in Ex group also raised significantly compared to Con group, while SYP, synapse plasticity, and Bcl-2 levels in Ex group were significantly lower than those in Con group. These indexes were back to normal in MUS group. BIC group had the highest levels of [Ca2+]i, astrogliosis, TUNEL positive cell, GFAP, Bax, caspase-3, caspase-12 cleavage, CHOP, and p-JNK, it also gained the lowest SYP, synapse plasticity, and Bcl-2 levels among all groups. Water maze test showed that Ex group had longer escape latency (EL) and less quadrant dwell time than Con group; all indexes between MUS and Con groups had no significant differences; BIC had the longest EL and least quadrant dwell time among all groups.</p><p><b>CONCLUSIONS</b>Activation of GABAA R could prevent intense exercise-induced synapses damage, excessive apoptosis, and dysfunction of hippocampus.</p>

ABSTRACT

<p><b>BACKGROUND</b>Intense exercise can cause injury and apoptosis, but few studies have reported its effect on the central nervous system (CNS). The initial reason for hippocampus injury is the excitotoxicity of glutamate and calcium overload. Intracellular free Ca(2+) ([Ca(2+)]i) overload may trigger the apoptosis pathway and neuron damage. The aim of this study was to investigate whether intense exercise could cause hippocampus apoptosis and neuron damage and then to determine which pathway was activated by this apoptosis.</p><p><b>METHODS</b>We used one bout of swimming exhaustion rats as models. Intracellular [Ca(2+)]i was measured to estimate the calcium overload by Fura-2/AM immediately after exhaustion; glial fibrillary acidic protein (GFAP) and synaptophysin (SYP) immunofluorescence were performed for estimating astrocyte activation and synapse plasticity 24 hours after exhaustion. Apoptosis cells were displayed using dUTP nick end labelling (TUNEL) stain; endoplasmic reticulum (ER) stress-induced apoptosis pathway and mitochondrial apoptosis pathway were synchronously detected by Western blotting.</p><p><b>RESULTS</b>An increasing level of intracellular [Ca(2+)]i (P < 0.01) was found in the hippocampus immediately after exhaustion. GFAP and SYP immunofluorescence showed that the astrocytes are activated, and the synapse plasticity collapsed significantly 24 hours after exhaustion. TUNEL stain showed that the number of apoptosis cells were notably raised (P < 0.01); Western blotting of the apoptosis pathway showed increasing levels of caspase-3 cleavage (P < 0.01), Bax (P < 0.01), caspase-12 cleavage (P < 0.01), C/EBP-homologous protein (CHOP) (P < 0.01), and phospho-Junaminoterminal kinases (p-JNK; P < 0.01) and decreasing level of Bcl-2 (P < 0.01). Our results proved that exhaustion can induce hippocampus injury and apoptosis by [Ca(2+)]i overload, with collapsed synaptic plasticity as the injury pattern and ER stress-induced apoptosis as the activated pathway.</p><p><b>CONCLUSION</b>Intense exercise can cause excessive apoptosis and synapse plasticity damage in the hippocampus with [Ca(2+)]i overload as the initial reason, and thus provides leads for therapeutic interventions in the brain health of athletes.</p>

ABSTRACT

This study aims to assess if adverse drug reactions (ADRs) to Kudiezi injection are allergic in origin. Hospital information system (HIS) data from 18 hospitals in China were used to carry out a nested case control design study. Included were patients who received dexamethasone for suspected allergic reactions after receiving Kudiezi injection. These were compared with non-allergic reaction people. Single factor logistic regression and multiple factor logistic regression were used to analyze data. Condition on admission, allergic history, dosage, disease status and drug combinations were taken into account in cases of suspected allergic reactions. After analysis in two subgroups we found that the condition on solvents had a significant effect, P values were 0.005 5 and < 0.000 1 on suspected cases of allergic reactions. For the first subgroup analysis, we found using other eight injections at the same time as Kudiezi injection could be risk factors in suspected cases of allergic reactions. For the second subgroup analysis combining using mannitol or fructose could increase risks. Based on this current research, condition on admission as well as the concomitant use of some other drugs may be the risk factors in suspected cases of allergic reactions. However, further research for verification is required. This study can provide guidance for safe clinical practice in using Kudiezi injection.

ABSTRACT

This study aims to explore the impact on patient's liver and kidney function by different dosage of Kudiezi injection. This study retrospectively analyzed 15 228 patients' records from 18 nationwide general hospital information system (HIS). All patients were treated with Kudiezi injection, 1 956 patients that were given doses of > 40 mL, which is above the recommended dose, acted as the observation group. Fifty-five patients receiving the recommended dose of < 40 mL were the control group. Data about alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine (Cr) and blood urea nitrogen (BUN) were collected before and after using Kudiezi injection, changes after treatment were outcomes. Also recorded were: age, costs, length of hospitalization and the patients' condition on admission. Propensity score method was used to balance 71 confounding variables such as gender, age, mortality, and costs. There were no significant difference on the four indexes between the two groups. It is hard to conclude that the use of Kudiezi injection over the recommended dose could influence the four indexes of liver and kidney from this data analysis. More conclusive evidence should be collected by further prospective study.
